Introduction

Introducing a new line to in vitro culture is done by either acquisition of material through collecting missions or transfer/donation, or from an existing accession in genebank's own field collection.

External acquisition

Follow the acquisition guide for material coming from outside the genebank. This will track the acquired material as new accessions.

Introduction from existing germplasm

This option is useful when the source plant already exists in the genebank (e.g. in a field collection) and the new line will be tracked under the same accession number.

The first step is to obtain the explants for initiation into in vitro from the source plant (i.e. nodal segments, apical meristems, roots, cotyledons, embryos, leaf discs, leaf blades, pedicles, petioles, anther, ovaries, etc.), followed by registering it in the database:

  1. Open In vitro > Introduction and scan the barcode of the source plant
  2. Select the number of new inventories (new lines) to be registered

  1. Select the maintenance policy and enter the quantity of each new inventory
    • Culture media, type of container, etc. can also be specified

  1. Click Preview and Save
    • Barcode labels for the new inventories will be automatically prepared

The lines are registered as new inventories and have their Propagation date set to "today". From this moment on, the lines can be monitored and subcultured as described in corresponding sections of this manual.

Naming convention

The accession number will be used as the inventory prefix and the next number in the sequence will be automatically assigned.

In the example above we took explants from FIELD 1 PL of accession GBK 121 and the introduced lines are called GBK 121 1 IV and GBK 121 2 IV:

When GBK 121 1 IV is subcultured, the lines will be called GBK 121.1 1 IV and GBK 121.1 2 IV as described in multiplication.
