Monitoring and disposal
Regular monitoring is carried out to detect and remove those in vitro cultures that exhibit any variation from whole plantlets, including somaclonal variation, contamination, hyperhydration, etc.
Samples should also be regularly monitored for signs of necrosis.
The types of variation the genebank will monitor are registered as Inventory action types with prefix IV_CTMN_
.
The monitoring and eventual discarding of material are two separate steps in GGCE.
Monitoring
- Identify the inventories that show variation and set them aside
- Open In vitro > Monitoring and scan the first inventory
- Select the type of contamination observed and hit Save
This creates the first entry in the list. Continue scanning the barcodes of inventories that show the same type of contamination, then hit Save.
GGCE will log these observations as Inventory actions and will not automatically discard or reduce the quantity on hand of the material. This allows you to review the observations and dispose only of material that meets your criteria.
Discarding material
After you have selected the material that requries disposal you can use In vitro > Discard.
- Set aside the material to discard
- Open In vitro > Discard
- Scan each inventory exactly once
- Each individual inventory is listed once in the table.
- The quantity to discard is increased by
1
unit for every barcode scanned.
- Review the list of material to discard
- Pay special attention to any line where the remaining quantity would be zero!
- Hit Discard material to reduce the available quantity on hand
- A final prompt is shown to confirm the action!
GGCE will reduce the quantity on hand of the discarded material and log the Discard inventory action.
